Development and characterization of microsatellite markers for blackfin tuna (Thunnus atlanticus) with the use of Illumina paired-end sequencing ArchiMer
Antoni, Luca; Luque, Patricia L.; Naghshpour, Kaylie; Reynal, Lionel; Saillant, Eric.
Twenty homologous microsatellite markers, or simple sequence repeats (SSRs), were developed for blackfin tuna (Thunnus atlanticus) through the use of a direct Seq-to-SSR approach. The number of alleles per locus ranged between 5 and 26, and the expected heterozygosity ranged between 0.640 and 0.969. Three loci displayed significant departure from Hardy-Weinberg equilibrium expectations, likely reflecting occurrence of null alleles. Another locus showed consecutive alleles that differed by one base pair only. Consequently, this locus may be prone to elevated rates of scoring errors. The remaining 16 loci will be valuable for studies in conservation genetics of blackfin tuna.

Effects of hypoxia and elevated ammonia concentration on the viability of red snapper embryos and early larvae ArchiMer
Bardon-albaret, Agnes; Saillant, Eric.
The effects of hypoxic conditions and elevated ammonia concentrations on the viability of embryos and newly hatched larvae of the red snapper (Lutjanus campechanus) were investigated. In all experiments, tested levels of hypoxia or ammonia concentrations were applied to embryos and unfed newly hatched larvae from three different spawns. Exposures began at 1 h post fertilization (pf) and lasted until all individuals in a group had expired. Survival rates were monitored daily in duplicates for each spawn in each treatment. Fertilized eggs exposed to 2 mg L− 1 dissolved oxygen (29% saturation) showed complete mortality before hatch while 81% of embryos in control groups (> 85% saturation) hatched and subsequently maintained high survival until 5 days pf....

Effets des conditions d'élevage sur la différenciation du sexe chez le bar, Dicentrarchus labrax, caractérisation du dimorphisme sexuel de croissance ArchiMer
Saillant, Eric.
The objective of this work was to describe gonadogenesis and sexual growth dimorphism in cultured sea bass Dicentrarchus labrax and to study sex determination in this species. Gonadogenesis was studied at the histological level in a population constituted of 30 sibs of sea bass. Sexual differentiation occurred between 125 and 416 days post-hatching (dph) (individual weights between 2.6 and 30g). 93% of the fish differentiated as males, 59% of these showing intratesticular oocytes. 30% of the males, among the biggest, differentiated early [165 days post hatching (dph)] and started spermatogenesis in their first year of life.  Ovarian differentiation also occurred early (165 dph) and females were heavier than males from 188 dph. The evolution of the...

Estimates of heritability and genotype-environment interactions for body weight in sea bass (Dicentrarchus labrax L.) raised under communal rearing conditions ArchiMer
Saillant, Eric; Dupont Nivet, Mathilde; Haffray, Pierrick; Chatain, Beatrice.
Genetic parameters for somatic growth rate in sea bass were estimated in two experiments where 27 (year-class 1997) to 30 (year-class 1998) families of sea bass (3 dams mated with 9-10 sires according to a full factorial design) were raised mixed in the same tanks starting shortly before hatching (48 h post fertilization). Trials were performed under two temperature (experiment 1, 1997: group high temperature (HT) and group low temperature (LT)) and two density regimes (experiment 2, 1998: group high density (HD) and group low density (LD)) with a subset of the mating design (18 families) being reproduced in the two experiments. All the fish were genotyped at 3-6 microsatellite loci which allowed them to be assigned to the individual breeders used in the...

Genetic variation for carcass quality traits in cultured sea bass (Dicentrarchus labrax) ArchiMer
Saillant, Eric; Dupont-nivet, Mathilde; Sabourault, Marie; Haffray, Pierrick; Laureau, Stanislas; Vidal, Marie-odile; Chatain, Beatrice.
Genetic parameters for carcass quality traits were estimated in 27 families of sea bass (3 dams x 9 sires factorial mating design), raised mixed in the same tanks starting before hatching. O. spring parentage was determined a posteriori using 6 microsatellite loci. Carcass quality traits were recorded at 818 days post fertilization (mean standard length: 32.6 +/- 3.1 cm). Genetic parameters were estimated from the sire half sib variance and covariance components. Heritability of body weight (BW) and carcass processing traits (standardized to body weight) percent head weight (Head%), percent viscera weight (Viscera%) and percent visceral fat weight (VisceFat%) were relatively high ranging from 0.48 +/- 0.15 (Viscera%) to 0.87 +/- 0.23 (Head%); the estimate...
